Totally depends on you! If you're the sort of person who enjoys a constant stream of new challenges, go for it! If you're the sort of person who views work as a means to living and not a place where you want to be over-taxed, then you can always decline.

I'm of the first sort, myself, and opportunities like promotions don't come along every day. That said, if you're in a comfortable place in your career, there's absolutely nothing wrong with turning down new responsibility.
